General Considerations
Pepsin is an acidic protein with a pI of 1.0. It is unstable above pH 6.9

Below pH 6, pepsin preferentially cleaves on the carboxyl side of L-phenylalanine, L-leucine, or L-tyrosine, where the amino side residue is preferably, but not limited to, an amino acid containing a hydrophobic side chain.
Pepsin is very useful in the preparation of immunoglobulin fragments, specifically from IgG and IgM class antibodies. With IgG antibodies, pepsin will yield F(ab’)2 fragments, whereas with most IgM molecules, it is possible to generate F(ab’)2, Fab, and Fv fragments by using the appropriate conditions.

Enzyme digestion varies with proteins as well as the operating conditions such as temperatureflow rate and pH.

By controlling these variables, it is possible to control the extent of digestion.
